The Localization of Skin-sensitizing Antibody in the Sera of Ragweed-sensitive Individuals by Electrophoresis
نویسندگان
چکیده
Starch electrophoresis was used to separate allergic sera into electrophoretically distinct albumin, alpha-, beta-, and gamma-globulin fractions. The identity of the various fractions was established by free and paper electrophoresis. The gamma-globulin fractions were also characterized by ultracentrifugation. The serum skin-sensitizing antibody to ragweed pollen was shown to be associated with the gamma-globulins in fourteen allergic sera by means of passive transfer technique. The mobility of the skin-sensitizing antibody corresponded to that of gamma-1 and/or gamma-2 globulins. All the other serum fractions were devoid of reaginic activity. Reaginic activity per se was not directly related to protein concentration. The discrepancies between these results, and those obtained by others and ourselves previously are discussed.

1. The removal of the beta(2)A-globulins from three sera from treated ragweed-sensitive individuals by immune absorption was associated with the loss of all detectable skin-sensitizing antibody activity as demonstrated by Prausnitz-Küstner testing. 2. Gel filtration studies, with sephadex G-200, indicated that the fractions containing only macroglobulins were devoid of all detectable skin-sensi...
متن کاملDetection of "non-precipitating" Antibodies in Sera of Individuals Allergic to Ragweed Pollen by an in Vitro Method
A hemagglutination test capable of demonstrating antibodies in sera of ragweed-sensitive individuals has been described in detail. This test involves coupling of the antigen to rabbit erythrocytes via stable azo bonds. The antigen-coated cells are then suspended in the serum which is suspected to contain the homologous antibodies. The test has been shown to be specific and highly sensitive. Pre...
متن کاملA Sensitive Neutralization Assay for Influenza C Viruses Based on the Acetylesterase Activity HEF Glycoprotein
Influenza C virus possesses specific neuraminate-O-acetylesterase as a receptor-destroying function. This enzymatic activity of the viral glycoprotein HEF (Hemagglutinin, esterase activity and fusion factor) can be visualized in situ by the use of distinct color substrates. Hereby the localization, as well as the quantity of synthesized HEF protein is detectable. We further developed the estera...
متن کاملThe Reaction of Placental GRP78 Protein with Sera from Women with Multiple Sclerosis
Background: Multiple Sclerosis (MS) with four different types is one of the well studied autoimmune diseases of the central nervous system. Generally, two-thirds of MS patients are females who are at risk of pregnancy-related complications. Inappropriate responses of mother’s immune system, such as antibody production against placental proteins, may lead to pregnancy-related disorders. The as...
متن کاملThe Sedimentation Properties of the Skin-sensitizing Antibodies of Ragweed-sensitive Patients
The sedimentation coefficients of the skin-sensitizing antibodies to ragweed were evaluated by the moving partition cell method and the sucrose density gradient method. The most reliable results were obtained by sucrose density gradient ultracentrifugation which showed that the major portion of skin-sensitizing antibodies to ragweed sediment with an average value of 7.7S (7.4 to 7.9S). This is ...
